Standard Operating Procedures (SOPs) – Best Practices for Optogenetic Experiments

Title: Best Practices for Optogenetic Experiments
Author: MouseTRAP
Date: 01/10/23
Version: 1
Abstract: Please follow this section if your experiment will be conducted in a MouseTRAP facility and requires you to construct an optogenetic probe with raw materials. Failure to follow the instructions of this section may result in probes with poor light transmission efficiency, which will negatively impact the outcome of your study. At the end of this document, you’ll find a list of recommended supplies. For non-MouseTRAP users, this document serves as a guideline of suggested procedures and practices to prepare your experiments in optogenetics.

Version Notes

[DATE]: Version [#]

  • Adjusted the After reward pause from 5s to 10s
    • Reasoning is explained at the end of the SOP, but was based on fibre photometry recordings that demonstrated mice needed more time to baseline
  • Included an additional formula for more data we can analyze: Premature Responses
  • Added another method to run the stimulus duration probe
  • Clarified details about the Inter Trial Interval
    • Specifically, we made it clearer that the ITI is variable, rather than a constant duration between each trial
